Cell Viability and Proliferation Assay

The proliferation capacity of Advancexo was assessed using a CCK-8 Assay kit on Human Dermal Fibroblasts. Approximately 2 x 104 cells were seeded to each well of a 6-well plate. Negative control was not treated, cells treated with a commercially available fibroblast growth factor (6µg/ml) were used as a positive control. The proliferation capacity was assessed, after trypsinizing and counting cells on the seventh day of expansion. The health of the human dermal fibroblasts post-treatment is displayed below.

Cytokine Profiling of Advancexo

The cytotoxicity of Advancexo, using the MTT Tetrazolium Assay was evaluated by the Institute of Industrial Research & Toxicology (IIRT). The study was conducted in compliance with the ISO 10993-5; 2009 (E) guidelines for biological evaluation of medical devices. The mouse fibroblasts L929 cell line was grown as monolayer cultures, maintained with Minimum Essential Medium supplemented with heat-inactivated FBS 10% and 2mM L-Glutamine. In Vitro Cytotoxicity Analysis

Advancexo’s cytokine profile was assessed with the help of a Multiplex Assay. The conditioned medium was collected for quantitative estimation of the presence of growth factors on the seventh day of culture. The control was obtained at day zero for baseline value. Advancexo showed several collagen-promoting and anti-inflammatory cytokines.
